US 378 has been around only since 1952, and apparently its endpoints have remained the same ever since. The west terminus is in Washington GA, at its junction with its parent, US 78:

That's looking west on US 378, or Lincolnton Road. The upcoming intersection is Sam McGill Parkway. In the distance is an "End" assembly, and behind the brown sign is another green sign. The "End" sign is fairly new...

Ivy/Nitzman, Oct. 2010

...but the green directional sign has been around longer:

Dawson, Sep. 2002

Note the old "End" assembly made use of a three-digit blank; a closer shot is below:

Today westbound US 78 is to the right, but originally it went straight ahead through downtown Washington on Robert Toombs Avenue - that's Business 78 now. Heading the opposite way on Business 78 (east on Toombs), the beginning of US 378 is signed thus:

Ivy/Nitzman, Oct. 2010

The ground-level assembly at lower right includes an "End Business 78" sign. Today eastbound US 78 approaches that intersection from the left, so the beginning of US 378 is a left turn:

Here's a view from the opposite direction (westbound US 78, or north on Sam McGill):

If you take that right turn, the first eastbound confirming marker used to look like this...

Dawson, Sep. 2002

...but I'm guessing that assembly has been replaced by now.

About 235 miles ahead, you reach the east end of US 378 in Conway SC. The last eastbound sign is posted before you reach Church Street (US 501/US 701) and 4th Avenue (old US 701, now SC 905):

Prince, 2000

Beyond there, US 378 is not well-marked, even though it curves east on 3rd and continues for another half-mile or so. The designation ends at Main Street, which was once mainline US 501, but today carries Business 501:

Nelson, June 2011

There was no "End" sign - nor any junction signage at all - just this green directional sign, posted underneath one of Conway's many lovely live oak trees. The same tree is visible in the shot below:

Google Maps Street View, 2008

That's looking north on Main; the east beginning of US 378 is to the left on 3rd. If you correctly guess that's the direction you need to go, you'll soon be rewarded with a sighting of the first westbound marker:
